ABSTRACT:

This study examine the dose of Chlorpromazine, an anti-psychotic drug that will induce catalepsy a parkinsonian symptom and also examine the effect of caffeine, a competitive adenosine A2A receptor antagonist on chlorpromazine induced parkinsonian symptoms in Wistar Rats of average weight 155g used in this study. It was focused on the hypothesis that blockade of adenosine A2A receptor could enhance catecholamine release in the basal ganglia and that caffeine has valuable neuroprotective effect against anti-psychotic drug induced parkinsonian symptom. Rats were divided into six groups of five animals each, group I animals received normal saline(0.9%Nacl) administered intraperitoneally(ip) , while other groups received 0.1mg/kg, - 0.8mg/kg chlorpromazine . In some experiments, animal received i.p concomitant administration 0.4mg/kg and 0.6mg/kg chlorpromazine and 10mg/kg and 15mg/kg caffeine while others received 10mg/kg and 15mg/kg caffeine pre-treatment with 0.4mg/kg and 0.6mg/kg chlorpromazine. Results obtained showed that pre-treatment administration(ip) with caffeine 15mg/kg significantly alleviated chlorpromazine induced catalepsy. The result seems to support the neuroprotective effect of caffeine in the concentration used in this study.
